Transferase activators boost enzyme function by promoting substrate binding, increasing reaction velocity, or alleviating autoinhibition. Kinase activators, for example, modulate phosphorylation cascades critical for cell signalling, and methyltransferase activators regulate epigenetic modifications affecting gene expression.

Dysregulated transferase activity contributes to metabolic disorders, neurodegenerative diseases, and immune dysfunction. For instance, impaired glycosyltransferase function can affect glycoprotein synthesis, leading to congenital disorders, while aberrant histone acetylation is implicated in neurodegeneration and inflammatory diseases.

Conversely, transferase activators aid therapeutic efforts towards metabolic regulation, neuroprotection, and immune modulation. Acetyltransferase activators may improve cognitive function by modulating histone acetylation, while glycosyltransferase activators could enhance mucin production for protective barrier functions in gastrointestinal diseases. Additionally, phase II metabolism enzymes, such as UDP-glucuronosyltransferases (UGTs), play a vital role in drug detoxification and xenobiotic clearance, where their activation could improve pharmacokinetics. Recent advances in structural biology, high-throughput screening, and computational drug design are refining the discovery of selective transferase activators.
