Consistent production of antibodies, specific to a single antigen, has enabled the practical use of antibodies as medicines and research tools. In the 1970s, methods using human-mouse hybrid cells enabled the production of identical antibodies – monoclonal antibodies (or mAbs). Initial medications produced in this manner tended to activate an immune response, and subsequent developments have produced engineered antibodies – notably humanised, then fully-human antibodies – which reduce unwanted immune responses. Recently biparatopic antibodies (bpAbs) have been developed, binding two distinct epitopes on the same antigen, creating even great affinity and specificity and opening new mechanisms of action.
Similarly, bispecific antibodies (bsAbs) combine binding sites for two antigens. Commonly used to bind cancerous cells and immune cells simultaneously – triggering an immune response to destroy the cancer cell. This mechanism requires far fewer bound antibodies to trigger cell death, so weakly expressed antigens can be targeted, and dosages can be far lower. Trispecific antibodies have now been developed, that further boost the immune cell response. Alternatively, bsAbs can be used to target multiple proteins associated with specific signalling pathways – preventing cancer cells working around the antibody through parallel pathways and reducing the chances of tumours developing resistance.
